چکیده

I t has been reported by Maxted (1) and Krause (2) that lysates of Group C streptococci infected with C1 bacteriophage contained an enzyme that had the ability to lyse Groups A, C, and E streptococci and their isolated cell walls. Crude and partially purified preparations of this enzyme were found to be useful for the isolation of Group A streptococcal cell wall components (M protein and C carbohydrate) (3-6), as well as for the preparation of cell wall-free protoplasts and protoplast membranes (7-11).

Identification of a lysin associated with a bacteriophage (A25) virulent for group A streptococci.

A phage-associated lysin was found in culture lysates resulting from the propagation of virulent bacteriophage A25 on the group A streptococcal strain designated K56. In contrast to the previously described group C streptococcal phage-associated lysins, A25 phage-associated lysin was more active on chloroform-treated cells, was not phage bound, and was active on some group G and H strains, as w...

Preparation of protoplasts of group H streptococci (Streptococcus sanguis).

Stable protoplasts of several strains of group H streptococci (Streptococcus sanguis) can be prepared by use of group C streptococcal phage-associated lysin in the presence of 30% raffinose. Sucrose cannot be substituted for raffinose. Protoplast formation did not require the addition of Mg2+; however, this cation enhanced their stability. Some other strains, also presumptive group H streptococ...

Purification and Physical Properties of Group C Streptococcal Phage-associated Lysin

A purification procedure for the Group C phage-associated lysin is described utilizing tetrathionate to protect the enzyme's -SH group(s) from thiol-inactivating agents. A 652-fold purification has been accomplished yielding a solution in which the enzyme activity corresponds to essentially a single band on polyacrylamide gel which accounts for 70% of the total protein in the preparation. A mol...

Preparation and Antigenicity of M Protein Released from Group A, Type 1 Streptococcal Cell Walls by Phage-associated Lysin

The lysis of Group A type 1 streptococcal cell walls by phage-associated lysin has been described. In the preparation of lysin, a new semisynthetic non-protein media to support growth of the propagating strain of Group C streptococci was employed. Following lysis of the cell walls, the resulting digest was partially purified by ion-exchange chromatography and ammonium sulfate precipitation. In ...

Studies on the Bacteriophages of Hemolytic Streptococci

The lysis of cell walls of hemolytic streptococci by a phage-associated lysin has been described. A method is presented for preparing the lysin from Group C streptococcal phage lysates. Following lysis almost all of the cell wall carbohydrate is recovered in solution. This material has the serological reactivity, physical-chemical properties, and values for nitrogen, rhamnose, and glucosamine s...
